What Is Theacrine?

Theacrine is a purine alkaloid found in Camellia kucha, a wild tea plant. Its chemical name is 1,3,7,9-tetramethyluric acid, and its structure is related to caffeine. It is now used in some energy and pre-workout supplements.

Research has linked theacrine to adenosine and dopamine signalling, both of which influence sleep pressure, motivation and arousal. Its effects are not fully settled.

Some studies describe stimulant-like effects, including increased locomotor activity and reduced fatigue. Others report sedative or sleep-promoting effects in animals. The 2020 review of theacrine research identifies these inconsistencies and notes that several mechanisms remain unresolved.

What Is Modafinil?

Modafinil is a prescription wakefulness-promoting drug. In the United States, it is approved for excessive sleepiness associated with narcolepsy, obstructive sleep apnoea and shift work disorder. In obstructive sleep apnoea, it treats residual sleepiness rather than the airway obstruction itself.

The usual adult dose is 200 mg once daily. It is generally taken in the morning for narcolepsy or obstructive sleep apnoea, or about one hour before a work shift for shift work disorder. Its effective elimination half-life is about 15 hours.

Modafinil inhibits dopamine reuptake and also appears to affect orexin, histamine and glutamate pathways involved in wakefulness. Its exact mechanism remains incomplete, but its clinical use and pharmacology are better defined than those of theacrine.

Does Theacrine Improve Energy and Alertness?

One of the most relevant human studies included 20 healthy men and women between 18 and 31 years old. Each participant received placebo, 150 mg of caffeine, or a multi-ingredient supplement containing theacrine and 150 mg of caffeine on separate test days.

The theacrine-containing product produced more favourable ratings for some subjective outcomes. Participants reported less grogginess, and the results showed trends toward greater attention, alertness, focus and energy.

The study did not test isolated theacrine. The supplement also contained white willow bark, SAMe, Rauwolfia vomitoria extract, citrus bioflavonoids and vitamin B12, and the exact amount of theacrine was not disclosed. The findings therefore cannot show how much of the effect came from theacrine rather than caffeine or the other ingredients.

Does Theacrine Improve Cognitive Performance?

The same trial measured reaction time and performance on the Trail Making Test and Digit Symbol Substitution Test. None of the three conditions produced a statistically significant improvement in these objective outcomes.

Participants could feel more alert without completing the cognitive tests more accurately or quickly.

The wider literature contains some positive findings, including modest improvements in selected cognitive measures when theacrine was combined with caffeine or used in pre-workout formulations. Study designs, doses and product compositions varied, and several trials involved small samples or short testing periods.

Taken together, the research does not establish a reliable cognitive-enhancing effect from theacrine alone.

How Strong Is the Evidence for Theacrine?

Much of the research on theacrine comes from animal or laboratory studies.

Animal experiments have examined possible effects on inflammation, pain, locomotor activity, fatigue, sleep, mood and memory. In mice, oral theacrine produced dose-related anti-inflammatory and analgesic effects in several experimental models. These results confirm biological activity, but they cannot determine whether humans experience comparable effects.

The 2020 review also summarised human studies reporting improvements in energy, fatigue, mood or concentration. Some tested theacrine with caffeine, while others used multi-ingredient supplements. The same review included studies that found no significant improvement in cognition or exercise performance.

The main weaknesses are small samples, short follow-up periods, inconsistent formulations and limited research on isolated theacrine.

How Do Their Mechanisms Differ?

Theacrine and modafinil both interact with systems involved in arousal, but they do so through different and incompletely understood pharmacology.

Theacrine has been linked to adenosine and dopamine signalling. Some animal studies describe adenosine receptor antagonism and increased locomotor activity. Others report sleep-promoting effects involving adenosine pathways.

Modafinil inhibits dopamine reuptake and increases extracellular dopamine in some brain regions. Its wake-promoting effects also appear to involve orexin, histamine and glutamate signalling.

Overlap in one neurotransmitter system does not make two compounds interchangeable. The magnitude, duration and clinical effects can still differ substantially.

Why Modafinil Has the Stronger Evidence Base

No direct head-to-head study appears in the supplied literature.

Modafinil has controlled clinical trials, established dosing guidance, approved indications and a documented adverse-effect profile. Theacrine has small human studies, supplement trials and a larger body of animal research.

Is Theacrine Safer Than Modafinil?

The 2020 review reported that daily supplementation for up to eight weeks did not produce clinically significant changes in the measured safety outcomes in healthy adults. It also described a four-week study of products containing theacrine and methylliberine that found no harmful changes in the monitored health measures.

These studies involved small groups, short follow-up periods and generally healthy participants. They cannot define rare adverse reactions or long-term risk.

A mouse study calculated an oral median lethal dose of 810.6 mg/kg, but animal toxicity figures do not establish a safe human dose.

Modafinil has a more detailed safety record. Common adverse reactions include headache, nausea, nervousness, anxiety, insomnia, dizziness and reduced appetite.

Its prescribing information also warns about serious skin reactions, angioedema, multiorgan hypersensitivity, psychiatric symptoms and cardiovascular events. Modafinil can affect drug metabolism and reduce the effectiveness of steroidal contraceptives.

A longer warning list does not automatically make modafinil less safe. It reflects broader clinical testing, formal prescribing requirements and more extensive adverse-event reporting.

Can You Take Theacrine With Modafinil?

The supplied sources contain no human study of theacrine and modafinil taken together.

Both affect alertness and arousal, so combining them could increase insomnia, anxiety, agitation, headache, palpitations or blood-pressure effects. This is a pharmacological concern rather than a documented interaction.

Modafinil already has an effective half-life of about 15 hours. Adding another stimulating supplement could extend unwanted effects, especially when taken later in the day.
